Once you make the payment, your public key gets added to the relay's database. After that anything you publish to the relay gets accepted. Posts/notes by pubkeys not on the relay DB will not get published to that relay, but you'll continue to see it from other public relays. Other public relays can pick up the note/post submitted to the paid relay if clients do the job of broadcasting it.
