Your ISP can inspect packets to see if you're using a VPN or Tor. This isn't difficult. They can't see what data the packets are protecting, but knowing you're using a VPN or Tor is obvious.

If your ISP knows further your public Twitter account and can correlate VPN or Tor packets with your public Twitter activity, then it's clear you're using the VPN or Tor to access Twitter.
