We say both really. Depends on the context
according to the OED the pronunciation changed early 19th century

The difference is primarily dependent on the region of the country one is from.
Route is pronounced as 'root' in the North East and 'rowt' can be heard predominately in the south. Pronunciation is very fluid, both are considered correct.
Okay, that's interesting. I guess hear "rowt" mostly from US media and movies.
